Cunningham war eine der ersten professionellen Fotografinnen in Amerika. Nach ihrem Fotografiestudium in Deutschland eröffnete Cunningham ein Porträtstudio in Seattle und fertigte allegorische Abzüge mit weichem Fokus in der Tradition des Piktorialismus an. 1932 trat sie einer Vereinigung modernistischer Fotografen bei, die unter dem Namen f/64 bekannt wurde und die malerischen Ideale des Piktorialismus ablehnte. Zu den Mitgliedern von f/64 gehörten Edward Weston, Ansel Adams und Willard van Dyke. Cunningham interessierte sich auch für menschliche Themen und fotografierte häufig die Hände von Musikern und Künstlern. Sie verkehrte zu verschiedenen Zeiten mit anderen ikonischen Fotografen des 20. Jahrhunderts, darunter Minor White und Dorothea Lange.

Cunninghams Arbeiten befinden sich in internationalen Sammlungen, wie u.a. im J. Paul Getty Museum, Los Angeles, USA; International Center of Photography, USA; Philadelphia Museum of Art, USA; Tokyo Metropolitan Museum of Photography, Japan; Museum of Modern Art, New York, USA; SFMOMA, San Francisco, USA.
